人外周血单个核细胞（PBMCs）的分离只需要一个标记步骤和两步分离过程。在第一个分离步骤中，红细胞聚集并沉淀。在第二步中，通过去除非pbmc（如中性粒细胞、嗜酸性粒细胞、血小板和残留红细胞）分离pbmc。

非pbmc用生物素偶联单克隆抗体和MACSprep™Anti-Biotin MicroBeads的混合物间接磁标记。通过将磁性标记的非pbmc保留在磁体中的MACS柱中来去除它们。

使用MACSprep™ PBMC Isolation Kit, human分离试剂盒、MACSmix™混匀仪和LS柱从10ml人edta抗凝全血中分离出未标记的PBMC。分离的细胞用CD45-VioBlue®、CD3-FITC、CD4-VioGreen™、CD8-APC-Vio® 770、CD14-APC、CD16-PE、CD56-PE、CD20-PE-Vio770和

7-AAD进行荧光染色，并使用MACSQuant®分析仪进行流式细胞术分析。基于CD45表达、散射信号和7-AAD，细胞碎片、非白细胞和死细胞被排除在分析之外。
